エンドセリンによるアポトーシス制御調節に関する細胞内情報伝達および分子機構

研究概要	申請者らはこれまでエンドセリンの病態生理学的役割、エンドセリンによる細胞増殖作用とその細胞内機序などを研究してきた。本研究により、エンドセリン-1などの血管作動物質が血管内皮細胞においてオートクリン/パラクリン・アポトーシス生存因子であることを見いだした。培養ラット血管内皮細胞、線維芽細胞を用い、種々の方法([i]NP-40lysis法によるnucleosomal ladderの検出[ii]抗single-stranded DNA抗体による免疫組織化学[iii]TUNEL法[iv]propedium iodide染色によるFACS解析)にてアポトーシスを評価した結果、以下の成績を得た。分化した動物細胞におけるジーンターゲティング法(promoterless neo法)により作成したc-myc欠失細胞株を用いて、c-myc遺伝子がpreproendothelin-1遺伝子を2相性に制御することを見出した(Endocrinology1997:138;4584-4590)。血清除去後アポトーシスは血管内皮細胞においてもc-myc依存性である。一酸化窒素などの血管拡張性因子も血管内皮・血管平滑筋におけるアポトーシスを惹起する。また、エンドセエリン、アドレノメデュリンは内皮細胞のアポトーシスを強力に抑制する(Circulation 1998 in press;Endocrinology1997;138:2615-2620)。エンドセリン‐1は血管内皮細胞における強力なオートクリン/パラクリン・アポトーシス生存因子である(Hypertesion1997:30;1198-1203)。その作用はET_B受容体を介する(J Cardiovasc Pharmacol 1998 in press)。
