チネイシンによる糸球体障害修復機構の研究

研究概要

1) 本年度は、前年度の成果の論文発表を行った(別刷り代支払)。2) ハブ毒で誘発した実験的糸球体腎炎モデルおよびテネイシン欠損マウスを用いて、テネイシンがメサンギウム細胞の増殖に必要なシグナル伝達を調整していることを示したが、正常マウスにハブ毒によって糸球体腎炎を誘発した後、テネイシン機能を中和する抗-テネイシン抗体を投与すると、対象群では時間経過とともに腎臓の機能が正常に戻るが、投与群ではテネイシン欠損マウスの場合と同様、腎臓の機能低下(アルブミン尿、血中尿素量の増加)が観察された。以上のことより、糸球体腎炎の修復過程においてテネイシンは、メサンギウム細胞の増殖に関連した重要な働きを持っていることが再確認された。
3) テネイシン欠損マウスから樹立したメサンギウム細胞の培養細胞株を用いて、テネイシンのPDGFやTGF-βの受容体発現に対する影響をPCR法を用いて解析したところ、テネイシンは、これらの受容体の発現調節に関与しているらしいことを示唆する結果を得た。現在は、この現象を更に詳細に検討中である。
