脊柱靭帯骨化症の発生に関与する骨増殖因子の解明と臨床応用のための基礎的研究

研究概要	平成9年度の研究で我々は、脊柱靭帯骨化症患者における靭帯細胞の異常が、いかなる細胞成長因子の関与で生じるかを、細胞生物学的、免疫組織学的および文し生物学的な手法を用いて解析した。細胞生物学的な解析から、OPLL症例の脊柱靭帯細胞に対して、BMP2が分化を、TGF-β1が基質合成を、bFGFが増殖を、そしてIGF-Iが増殖、基質合成を促進させることが明かとなった。また、細胞成長因子間の相互作用の検討から、BMP2の分化誘導作用は、IGF-Iによって促進的に、bFGFによって抑制的に調節を受けることが示された。免疫組織学的にOPLL症例の後縦靭帯組織におけるPCNA(組織増殖核抗原)の発現を検討した。その結果、OPLL症例では椎体隅角部のみなら椎体中央部の後縦靭帯組織においても、その構成細胞が高い増職能を獲得していることが示された。この靭帯細胞の性状異常がOPLLの発生進展に密接に関わっていることが推察された。 In situ hybridization法を用いて、骨形成過程におけるXI型コラーゲン遺伝子の発現を解析した。その結果、従来から軟骨組織特異的とされてきたXI型コラーゲン遺伝子が、非軟骨組織でも相当量発現していることが示された。今後は、OPLL症例の脊柱靭帯組織におけるXI型コラーゲンの発現・局在について、その詳細を解析する予定である。また、脊柱靭帯組織内におけrく、マトリックスおよび成長因子の相互作用につても検討を進めたい。
