ヒト子宮内膜の増殖抑制機序に関する分子生物学的研究

研究概要	正常内膜腺上皮は分泌期にプロゲステロン(P)の作用により増殖を停止し分泌能を獲得すると考えられている。今回の研究はPによる内膜腺上皮の増殖抑制機序を細胞周期調節因子の観点から研究するために、以下の実験をおこなった。(1)細胞増殖を促進する作用を有する各種サイクリン、サイクリン依存症燐酸化酵素(cdk)の発現を免疫染色によって観察したところ、増殖期ではその発現がみられたが分泌期では著明に低下した。また、細胞増殖を停止させる作用を有する癌抑制遺伝子産物の発現を免疫染色によって観察したところ、p27が分泌期腺上皮に強く観察された。(2)腺上皮を分離・培養しプロゲステロン(P)添加の有無とこれら因子の発現をWestern blottingによって調べたところ、Pの添加によってサイクリンおよびcdkの発現が低下するとともにPの濃度依存症にp27の発現が増強した。(3)正常内膜におけるp27mRNAの発現をNorthern blottingによって観察したところ、増殖期と分泌期の間で著明な差はみられなかった。(4)培養腺細胞に蛋白合成阻害剤であるcycloheximideを添加しその後のp27蛋白量をP添加の有無で経時的に比較したところ、P添加群の方が未添加群よりp27蛋白量が長く遺残した。(5)培養腺上皮細胞にp27発現ベクターを導入してp27蛋白を強発現させたところ、p27導入後に細胞増殖指標であるproliferating cell nuclear antigen(PCNA)が低下した。以上のことより、分泌期のPによる腺上皮細胞の増殖抑制にはPによってサイクリンおよびcdkの発現が抑制されることに加えて、Pによる半減期の延長によって誘導されたp27が増殖抑制に機能的に関与することが重要であると考えられた。
