卵巣癌集学治療の細胞生物学的polyclonalityへの影響の基礎的研究:抑制癌遺伝子p53および癌退縮遺伝子MAGEを中心として

研究概要

【今年度の進行状況】
1)DMBA-OC-1の酵母を用いたラットp53遺伝子の機能的変異検出法による結果
赤色コロニーの形成率100%で、DMBA-OC-1はp53のmutation率としては100%のクローンであることが証明された。さらに、mutation部位のsequenceによるとcodon212のCACがATCにmutationしており、hystidineがIleusineに変化していることが確認された。この部位が、発癌とどのような関係にあるかは、現在検討中である。
2)実験腫瘍(DMBA clipping法による)検索を計画中であり、平成9年12月および平成10年1月、3月の3度に各30(計90匹)において腫瘍の作成を試みている。腫瘍の形成は平成10年5、6月ごろになる予定であり、その時点での検索を予定している。
3)現在、当教で樹立した他のヒト卵巣癌および子宮体部癌について酵母コロニー形成法により検索中である。
4)卵巣癌組織で発現を確認したMAGE遺伝子のうちMAGE-4について血清中の測定系の確立により現在測定を行っている。また、他の非悪性腫瘍や正常人におけるMAGEのカットオフ値について検討を行った。
【今後の予定と考察】
1)これまで、検索の困難であったラットにおける卵巣癌のDMBAによる発癌機構にはp53のmutationが関係している可能性が示唆された。
2)DMBAにより卵巣には多彩な腫瘍を形成するため、その組織型との関連および、発現時期(p53)が腫瘍発生の時期との関連も含めて問題になるかもしれない。
3)2)に関連して、多段階発癌DMBAがinitiationとpromotionのどちらに作用しているかの確認も必要かもしれない。(皮膚癌等ではすでに解析されている)
4)ヒト癌の結果によっては、従来のPCR-SSCP法よりもさらに高感度かつ特異的であるため、発癌機構に大きな手掛かりを得る可能性がある。
5)MAGE遺伝子については腫瘍マーカーともなりうる可能性が示唆されている。
