口腔・顔面領域でのカルシウム結合蛋白質の分布とその機能の解明

研究概要

正常な細胞内にはセカンドメッセンジャーであるCa^<2+>と直接結合することにより構造変化を起こし、機能を発現するカルシウム結合蛋白質(CaBP)と呼ばれる蛋白質が存在している。本研究ではラット口腔・顔面領域におけるCaBPのうちcalbindin D28k(CB),calretinin(CR)およびSl00の分布や組織発生との関連を明らかにし、さらにCaBPと炭酸脱水酵素(CA)との関係を明らかにすることを目的とした。さらに、神経損傷による変化を解析し、口腔・顔面領域でのCaBPの機能の解析を試みた。
1) 味蕾におけるCaBPとCA IIの分布と発生
舌乳頭のうち有郭乳頭と葉状乳頭の一部の細胞にCBが認められ、それらはいわゆる味細胞(type III cells)であることが分かった。CB、CRのいずれも神線維にも認められた。一方CA IIは有郭乳頭と葉状乳頭の一部の細胞に認められ、これらは支持細胞(type I cells)であった。味蕾の発生および再生においてCB陽性細胞は味蕾が形成あるいは再生してから約10日後に認められた。このことはCBが味覚伝達のみならず味細胞の維持に関与している可能性が示唆された。CAIIは、味蕾が形成される2〜3日前に味覚上皮に認められた。この時期には小唾液腺である舌腺の発生が始まる時期であり、CA IIが味蕾形成のみならず舌腺の形成とも関与している可能性が示唆された。
2) 臼歯歯髄および切歯歯根膜Ruffini神経終末におけるCaBPとCA IIの分布
ラット臼歯歯髄にはCB陽性神経線維が認められ、それらが象牙前質に進入していることが明らかになった。また、S-100βはシュワン細胞以外にも軸索に認められた。ラット切歯歯根膜には機械受容器であるRuffini神経終末が豊富に分布することが知られている。このRuffini神経終末には今までに報告されているCaBP以外にCRやS100β、さらにCA IIがRuffini神経終末軸索終末に存在することが明らかになった。支配神経損傷後のRuffini神経終末再生過程においては、CRは再生過程の後期にが発現することが分かった。また、発生過程においても同様の所見が認められた。このことからCaBPが歯髄や歯根膜での感覚受容機構に重要な投割を担っていることが推察された。
