ラット骨芽細胞における細胞骨格遺伝子アクチンmRNAの細胞内分布及び輸送機構

研究概要

細胞内における各種mRNAの発現有無については最近多くの研究がなされているが、mRNAの細胞内局在についてはほとんど知られていない。そこで本研究は、beta-actin mRNA及びgamma-actin mRNAのprobeを用い、正常ラット頭蓋骨骨芽細胞における各種actin mRNAの細胞内局在についてIn situ hybridization法を用い検討した。実験に用いた培養細胞は、正常及び大理石骨病モデルラット(tl/tl)において、頭蓋骨より骨芽細胞を分離し初期培養によって得た。In situ hybridization法のための試料には、培養3,5日目のものを用いた。前処理後、50%formamideでincubateし、その後はbeta及びgamma-actin mRNA Probeを含むbuffer液にてhybridizationを行った。anti-digoxigenin mouse IgG-AP conjugateを使いincubate後、染色、封入後、光顕観察を行った。
(1)(昨年までの結果)正常骨芽細胞では、beta-actin mRNAは細胞突起、細胞膜周辺部、さらに細胞の核周辺一方向のみの細胞質部分にシグナルが観察され、gamma-actin mRNAは核周囲全周にシグナルが観察された。これらの結果から、正常骨芽細胞ではbeta-actin mRNAとgamma-actin mRNAとは各々細胞内局在部位が異なることが明らかになった。一方大理石骨病ラットの骨芽細胞では、beta-actin mRNA及びgamma-actin mRNA共に細胞内ではdiffuseに分布し、細胞内局在化が崩れていた(J.CellSci,111,1998)。csf-1投与後骨芽細胞のbeta及びgamma-actin mRNAの細胞内局在化についても検索を行った結果、betaとgamma-actinmRNAでは分布及び回復状況に差異が認められた。
(2)(本年度の結果)本年度はcsf-1投与後骨芽細胞のmRNA局在についての統計学処理と論文作成を行った。統計学的処理の結果、csf-1投与後大理石骨病モデルラットの培養骨芽細胞のbeta及びgamma-actin mRNAの局在化について検索した結果、beta-actin mRNAについてはcontrol群のmRNA局在部位に回復していた。一方gamma-actinmRNAについてはdiffuseで回復していなかった。以上の結果より大理石骨病モデルラットの骨芽細胞では、上記各mRNAの細胞内輸送機構において、csf-1がbeta及びgamma-actin mRNA輸送において異なる作用をもつ可能性が示唆された。
