歯周病原性細菌感染細胞アポトーシスにおけるIL-1β変換酵素の役割の解析

研究概要	線虫のced-3はアポトーシスに直接関わる遺伝子の一つである。そのホモログとして最初に同定されたがIL-1β変換酵素(IL-1β converting enzyme;ICE)である。その後、ICE/CED-3相同遺伝子が脊椎動物ではファミリーを形成していることが明らかとなった。現在では、ICE/CED-3ファミリーはカスペース(caspase)と総称されている。カスペースファミリーメンバーなかでも、caspase-1(ICE)とcaspase-3(CPP32/apopain)は哺乳動物細胞におけるアポトーシス制御タンパクとして注目されている。本研究では、A.actinomycetemcomitans感染マクロファージにアポトーシスが誘導される際に、ICE/caspase-1とCPP32/apopain/caspase-3が関与しているか否かを検討した。まず、それぞれの酵素の特異的阻害剤であるICE/caspase-1阻害剤(Z-VAD-FMK)とCPP32/apopain/caspase-3阻害剤(DEVD-FMK)を用いて、感染マクロファージの致死活性におよぼす影響を調べたところ、Z-VAD-FMKおよびDEVD-FMKを添加することにより著しい抑制効果が認められた。さらに、この抑制効果はアポトーシス細胞の検出系(フローサイトメトリーでの解析、DNAアガロース電気泳動、ヘキスト染色後の核の形態観察)でも確認された。次に、Z-VAD-FMKのICE阻害効果を確認する目的で感染マクロファージからのIL-1β産生の変化を調べた。A.actinomycetemcomitansを感染させたマクロファージからはIL-1βが産生されていたが、Z-VAD-FMKを添加することでIL-1βの産生が著しく抑制された。このことから、マクロファージにA.actinomycetemcomitansを感染させることで、細胞内のICE/caspase-1の酵素活性が亢進し、その結果アポトーシスの誘導とIL-1βの産生が引き起こされていることが示された。今後、ICE/caspase-1とCPP32/apopain/caspase-3の細胞内での酵素活性を測定して、2つのプロテアーゼの関与をより明確にしていくつもりである。
