ヒト口腔癌細胞の運動能に及ぼすEGFの影響と細胞内シグナル伝達の解析

研究概要

現在、保有するヒト口腔扁平上皮癌細胞株のうち、金コロイド法による運動能の変化を指標として得た、EGFに対する応答性が最も高いクローン(S-1株)を用い以下の検討を行った。
1) 酵素阻害による運動能の変化
a) EGF受容体チロシンキナーゼ阻害剤(Erbstatin)
b) PIターンオーバー阻害剤(Psi-tectorigenin)
c) PKC活性阻害剤(Calphostin C)
d) PI3キナーゼ阻害剤(Wortmanin)
2) EGF処理によるPKC活性の変化
共焦点レーザー顕微鏡によるPKCの細胞内トランスロケーションの観察
以上の結果、EGFがEGF受容体に結合後、受容体の細胞内ドメインの自己リン酸化が生じ、次いでPLCrの活性化を介したPKCの活性化が生じ、ヒト口腔癌細胞の運動能の亢進が起こることが明らかとなった。
また、この現象には、ErbB1受容体のホモ二量体形成のみならずErbB1とErbB3のへテロ二量体の形成が関与している可能性が示めされた。
