口腔顎顔面領域の間葉系再細胞内力学的刺激情報伝達系におけるMAPキナーゼの役割

研究概要	矯正治療を前提に便宜抜去されたヒト上顎第一小臼歯歯根膜よりoutgrowth法を用いて10%ウシ胎仔血清を含むD-MEM中で37℃、5%CO_2,95%Airの条件下で歯根膜細胞の培養を行った。細胞がコンフルエントになった時点でトリプシン-EDTAを用いて細胞をシャーレから剥がし、コラーゲンゲル中に包埋して三次元培養系に移した。その後、ゲルをシャーレから遊離させてメディウム中に浮遊させ、細胞を張力から解放させる刺激を加え、一定時間経過後、回収した培養上清をラット骨髄細胞培養系に加えたところ、活性型ビタミンD3存在下でTRAP陽性多核破骨細胞様細胞の形成を有意に促進した。さらに、これらの三次元培養された歯根膜細胞を継時的にRIPAbufferで処理して細胞内タンパクを抽出し、リン酸化チロシンを認識する抗体を用いたウェスタンブロット法を行ったところ、60kd付近のタンパクにチロシン残基のリン酸化の亢進が見られることが明らかとなり、さらにsrcに対するモノクローナル抗体を用いたウェスタンブロットを行ったところ、張力解放刺激に伴って一過性にsrcタンパクの発現が上昇することが確認された。さらに、各種インヒビターを用いてゲルの収縮への影響を検討したところ、チロシンキナーゼのインヒビターであるGenisteinが30μMの濃度でゲルの収縮を一部阻害することが明らかとなり、チロシンキナーゼを介する系が歯根膜細胞の力学的刺激情報伝達に少なくとも一部関与する可能性が示唆された。
