PolyamineとZ-DNAの相互作用についての構造科学的研究

研究概要	1999年にはZ-DNA認識蛋白質がZ-DNAとの複合体結晶でX-線結晶構造解析されその蛋白質がRNAaminaseであることがわかった。この蛋白質は核酸塩基のグアニン塩基をアデニン塩基に変換する酵素であることがわかり、それによって核酸のグアニン塩基がアデニン塩基に変換され本来目的とする蛋白質の塩基コードと違う塩基に変換されてしまうため転写異常を起こし目的外の蛋白質を作らせる働きがあることもわかってきた。このようなことが明らかになるにつれPolyamineとZ-DNAのなぞはより深くなってきた。高濃度のPolyamineとDNAを混ぜ合わせるとDNAは左巻きZ-DNAになることも明らかにされている。そこでわれわれは様々なPolyamineとDNAの相互作用様式を調べるため様々なPolyamineとZ-DNA複合体結晶構造を明らかにし、PolyamineのZ-DNAに対する作用を明らかにすると同時にZ-DNAを安定化するPolymineのデザインに成功した。そこでわれわれはこれらのPolymineをさらにデザインしさらにZ-DNAを強力に安定化すると考えられるPolyamineとZ-DNAとの複合体結晶を作成し、X-線結晶構造解析を行っていく予定である。さらにZ-DNAのなりやすさにシークエンス依存性があることなどもわかってきたためシークエンスとZ-DNAの関連性をX-線結晶構造解析法を使用して明らかにしていく予定である。分子動力学法を用いた計算機科学的研究ではZ-DNAは金属イオンによって安定化されさらにその金属の質量が大きくなるにつれて安定化することがわかってきた。またPolyamineなどのDNAに結合しやすい物質といっしょに分子動力学計算を行うと回りの金属イオンの質量が低くてもZ-DNAは安定化することがわかった。しかしPolyamine単体ではZ-DNAを安定化できないこともわかった。さらに分子動力学計算で(d(AC)_4/d(TG)_4)のシークエンスではどのような条件でもZ-DNAにならないことが明らかになった。
