植物油脂が動物行動に影響を与える機構の解明

研究概要	α‐リノレン酸制限-高リノール酸の紅花油食(Saf)を二世代にわたって与え、脳内ドコサヘキサエン酸(DHA)含量を半減させたラット及び高α‐リノレン酸紫蘇油食(Per)を与え、脳内DHAレベルを高く保ったラットについて、行動解析及び生化学的解析による比較を行い、次のことが明らかとなった。明度識別型学習能試験ではPer群に比べてSaf群で有意に正反応率が低かった。レバ-押し回数はPer群に比べてSaf群で有意に高く、二世代目よりSaf群にエイコサペンタエン酸エチルエステル(EPA)を添加し、脳内DHAレベルをPer群と同じレベルまで回復させた群では、レバ-押し回数はPer群のレベルまで低下した。また、Saf群ではPer群と比較して新規の環境に暴露したときの探索行動量が有意に高くなっていた。マイクロ波処理脳各部位の神経伝達物質レベルを測定した結果、ドーパミンやノルエピネフリン、セロトニンなどのモノアミン含量には有意な変化は見られなかった。一方、アセチルコリン含量は海馬でのみSaf群で有意に低下していた。今後、アセチルコリン代謝系を解析することでその機構を検討し、行動に及ぼす影響との関連性を解析する予定である。グルタミン酸受容体の結合活性を比較したところ、AMPA型グルタミン酸受容体結合活性が脳各部位においてSaf群で特異的に低下していた。NMDA型受容体やカイニン酸型受容体には差は見られなかった。AMPA受容体は、長期増強の過程で応答が増強されることが報告されており、この現象がSaf群の学習能低下と関連づけられるかどうかが今後の課題である。
