研究概要	α-リノレン酸(18:3n-3)欠乏-高リノール酸(18:2n-6)の紅花油(Saf)を2世代にわたって摂取し、脳内ドコサヘキサエン酸(DHA、22:6n-3)含量が半減したラットでは、高α-リノレン紫蘇油(Per)を与えたラットと比較して、明度識別型学習能試験の成績が低い。このような学習能低下が、離乳期以後のn-3系列脂肪酸の摂取で回復するかどうかを検討した。Saf群の2世代目離乳後より、DHAを与えたところ(Saf+DHA)、脳内DHA量はPer群のレベルまで回復し、Saf群で相補的に増加していたドコサペンタエン酸(DPA、22:5n-6)含量もPer群のレベルまで低下した。しかし、Saf+DHA群の学習能は、Saf群と同レベルでPer群と比較して有意に低かった。レバー押し回数など他のパラメータもSaf群と似た挙動を示し、その行動パターンはSaf群と類似していた。脳各部位の神経伝達物質含量は、ドーパミンやセロトニンなどのモノアミン含量は3群間で差はなかった。アセチルコリン含量は、前頭皮質及び海馬でSaf+DHA群では他の2群と比較して低い傾向を示したが、学習能とは相関しなかった。一方、脳内神経成長因子(NGF)含量は、海馬ではPer群と比較してSaf群で有意に低く、逆に梨状様皮質では有意に高かった。Saf群の2世代目離乳後より、エイコサペンタエン酸(EPA、20:5n-3)を与えたSaf+EPA群では、Saf+DHA群と同様に脳内DHA含量はPer群のレベルまで回復していたが、NGF含量はいずれの部位でもSaf群と同レベルであった。以上より、n-3系列脂肪酸欠乏油脂による学習能低下やNGF含量の変化は、胎生期から授乳期の脳の発達段階でのDHA含量の低下による不可逆的なものである可能性が示唆された。但し、DHA・DPA以外で、20:4n-6や22:4n-6など回復が不十分な脂肪酸も見られたことから、これらが関与している可能性も残されている。
