分子動力学的な構造類似性を考慮した低分子プロープによる細胞分化誘導の分子機構解析

研究概要

白色脂肪細胞において、β-作動薬によって脂肪酸の生成が誘導される。このときインスリンは、濃度依存的(0〜10nM)にこの脂肪酸生成を抑える。
分子設計合成したsteric hindered phenol(TX-1123)は白色脂肪細胞において、1.2x10^<-11>Mのインスリン存在下、濃度依存的にこのインスリン作用を抑制し脂肪酸生成を増大させた。一方2-nitroimidazole(TX-1877)は、濃度依存的にインスリン作用を促進し脂肪酸の遊離を促進した。TX-1123,TX-1877いずれの場合も、単独では脂肪酸生成系に対する効果はなく、インスリン存在時のみ協調的に作用を発現した。さらに、これら化合物は、糖代謝系に関わるHexokinase(HKI〜IV)およびGlucose Transporter(GLUT1〜4)の発現強度へは影響を示さなかったことから、インスリンの関わるこれらの糖代謝系への直接的作用は有さないと考えられた。TX-1877はモデル膜を安定化させたことから、細胞側のインスリン感受性の修飾が作用本体である可能性が示唆され、現在詳細な解析を行っている。
