研究概要	ゲノム・インプリンティングは、生殖細胞での性特異的なインプリントに応じて、染色体上の対立遺伝子が父親由来または母親由来で異なって発現する現象である。染色体DNAのメチル化・DNA複製・高次のクロマチン構造など、エピジェネティクに制御されている。ヒト15番染色体長腕(15q11-13)の異常によって発生するプラダー・ビリー症候群(PWS)とアンジェルマン症候群(AS)は、その成立にインプリンティングが関連した神経疾患・発生異常である。本研究では、small nuclear ribonucleoprotein-associated polypeptideN(以下、SNRPNと略する)遺伝子上流領域に想定されたインプリンティング・センターの構造と機能に注目して、インプリンティングの分子機構の解明を目指した。中でも、DNAのメチル化がインプリンティング機構に重要であることから、DNAのメチル化とメチル化CpG結合蛋白質の役割に焦点を置いて解析した。メチル化CpG結合蛋白質(PCM1)を新たに同定して、その機能を解析した。PCM1には少なくとも5つのスプライス型があり、分子中央部分の制御配列CXXCドメインが2または3コピーに変化した。PCM1は核内に多数のフォーカスを形成し、この局在はDNAメチル化に依存していた。PCM1はメチル化されたSNRPN遺伝子プロモーター(CpGアイランド)に特異的に作用して転写活性を抑制した。スプライス型によって転写制御が異なっており、2コピーのCXXCをもつPCM1はメチル化SNRPNプロモーターだけを抑制し、一方、3コピーのCXXCをもつPCM1はメチル化・非メチル化SNRPNプロモーターの両者を抑制した。メチル化CpG結合蛋白質の機能活性がCXXCで調節されていることは新知見である。このようにPCM1は、インプリンティングを含めた遺伝子発現の抑制、核内での染色体構造への関連性が示唆されている(日本人類遺伝学会第42、43回大会;論文投稿中)。
