遺伝子高チロジン血症における肝細胞アポトーシスおよび肝細胞癌の研究

研究概要	本研究では目的は遺伝性高チロジン血症における遺伝子発現異常と肝細胞死及び肝細胞癌の発生機序の解明、および肝細胞障害の遺伝子治療のモデルの確立である。昨年度の研究に引き続いて行った本年度の研究において、以下の事柄が明らかになった。遺伝子高チロジン血症の臓器障害モデルであるFAH-HPD欠損マウスにおいてHPDを発現する組み換えアデノウイルスあるいはチロシン代謝系の中間産物であるホモゲンチジン酸を投与すると、投予後6時間で肝細胞はアポトーシスに陥る。本年度の研究では(i)FAH欠損ではフマリルアセト酢酸が細胞内に蓄積すること、(ii)アポトーシスに先立って投予後1時間でミトコンドリアからチトクロームCが放出されていること、(iii)cell freeの系で精製フマリルアセト酢酸を正常対照マウスから調整したミトコンドリアとincubateすると、チトクロームCが放出されることなどが判明した。また、アポトーシス関連蛋白分解酵素租外在であるカスパーゼ阻害剤は(i)2重欠損由来初代培養肝細胞のアポトーシスをほぼ完全に防止する(ii)in vivoでアポトーシス誘導前に2重欠損マウスに投与すると、肝障害と個体死を防止できることが判明した。以上の結果から高チロジン血症I型ではチロシン分解系の中間代謝産物がミトコンドリアに直接作用し、細胞死を招いていることが考えられた。一方、この疾患では生存すると肝細胞癌が発生する。このモデル系において慢性肝障害を作成することで発がんの機序の解明も可能になるものと考え、ホモゲンチジン酸少量長期投与による肝障害モデルを作成した。現在、慢性肝障害の検討を進めているところである。この慢性肝障害もでるによって、遺伝子異常を補正した肝細胞が肝臓内で生存し増殖する課程の観察を可能にすることも期待される。
