CaCo-2cellにおけるアポA-IVの合成・分泌機序について

研究概要	アポA-IVはヒト小腸粘膜細胞において特異的に合成・分泌されるユニークなアポ蛋白である。申請者はこれまで主としてラットを用いてこのアポ蛋白の合成機序を明らかにしてきたが、今回さらにアポA-IVの生理的機能を明らかにする目的でヒト大腸ガン由来のCaCo-2 cell(ヒト小腸粘膜細胞の機能を保持)を用いて、脂肪酸の種類によるアポA-IV合成に与える影響について検討した。 CaCo-2 cellの細胞極性を保持した条件で実験するため、mambrane chamber付きのTranswell(Costar)を用いて検討した。細胞がconfluentになってからserum-free DMEMおよび2種類の脂肪酸を加えて比較した。0.6mMの中鎖脂肪酸であるCaprylic acid (C_<8:0>)と長鎖脂肪酸のOleic acid(C_<18:1>)を12mM Na taurocholate、2mM phosphatidyl cholineとともにculture wellに加えた。incubation timeは6および24時間とし、まずlower wellに分泌されるリポ蛋白について検討した。TGはC_<18:1>の24時間においてのみ有意に増加(2倍)していたが、アポA-IVmRNAレベルに変化は認められなかった。そこで脂肪酸の濃度を0.6mMから6.0mMに増加させて検討したところ、C_<18:1>の6および24時間においてC_<8:0>に比べ16〜20倍のTGの増加が認められた。しかしこの脂肪酸濃度においては細胞障害が著しく、mRNAを検討するに十分なRNAを得ることができなかった。そこで現在脂肪酸の至適濃度を決定するため種々の濃度で検討を行い、同時にmedium中と細胞内におけるアポA-IVの蛋白濃度も測定中である。この結果をふまえ、さらにアポB-48も測定してアポA-IVとの機能的差異を明らかにする予定である。
