CaCo-2cellにおけるアポA-IVの合成・分泌機序について

研究概要	ヒトアポA-IVは小腸粘膜細胞において脂肪の吸収時に特異的に合成され、リンパを経て血中に出現するアポ蛋白であるが、その機能に関しては十分に解明されていない。申請者はこれまで主としてラットを用いてこのアポ蛋白の合成機序について検討してきたが、今回さらにアポA-IVの合成機序を明らかにする目的でヒト大腸ガン由来のCaCo-2 cell(ヒト小腸粘膜細胞の機能を保持)を用いて、2種類の脂肪酸(oleic acidおよびcaprylic acid)によるアポA-IV合成に与える影響について検討した。 CaCo-2 cellの極性を保持するためmambrane chamber付きのTranswell(Costar)を用いて検討した。細胞がconfluentになってからserum-free DMEMおよび上記2種類の脂肪酸を加えて比較した。3.0mMの長鎖脂肪酸であるOleic acid(C_<18:1>)あるいは中鎖脂肪酸のCaprylic acid(C_<8:0>)に12mM Na taurocholate、2mM phosphatidyl cholineを混合し、ミセル溶液として culture well に加えた。培養時間は6および24時間とし、まずlower wellに分泌される中世脂肪量について検討した。C_<18:1>を負荷した場合TGは6時間で4倍、24時間で24倍増加し、C_<8:0>はそれぞれ1.3倍、10倍増加した。しかし24時間後の細胞は高濃度の脂肪酸のため細胞障害が著しく、mRNA量の測定が不可能であったため、6時間の培養のみについて検討した。6時間後の細胞内アポA-IVmRNAレベルをノーザンにて測定したところ、C_<18:1>はC_<8:0>に比べ有意に増加していた。また培養液中のアポA-IV濃度をウエスタンブロットにより検討したところ、C_<18:1>において培養液アポA-IV濃度はコントロールおよびC_<8:0>に比べ、有意に低下していた。これらの結果の解釈は難しいが、C_<8:0>はアポA-IVの細胞からの分泌を亢進するが合成には影響を与えず、これに対してC_<18:1>はアポA-IVの分泌にはむしろ抑制的に作用するが、transcriptionに正の影響を与えていることが明らかとなった。これらの作用の違いは脂肪酸の長さによるものと考えられた。
