ヒト細胞の紫外線誘導突然変異を制御するサイトカインとプロテアーゼに関する研究

研究概要

波長254nm 付近の紫外線でヒト細胞を照射すると、DNAの傷害とその修復反応、遺伝子突然変異さらにはそれに引き続く形質レベルでの突然変異の発生(変異誘導)、あるいは細胞分裂の停止など生死にかかわる様々な反応が起こる。本研究の目的は、紫外線応答の中でも特に突然変異の誘導に的を絞り、サイトカイン・プロテアーゼの変異誘導制御系路の骨格を明らかにすることである。
すでに樹立してある紫外線誘発プロテアーゼの活性レベルが高い派生株から高活性レベルの細胞溶解サンプルを得て、目的酵素を抽出した。粗酵素標品中のプロテアーゼ活性は、アンチパイン高感受性というこれまでの知見に加えて、ALLNalで増大するという新たな知見が得られ、比較分子量は約2万であることが確認された。変異誘導との関連については、まずK-ras codon 12塩基置換変異検出法であるPCRとDifferential dot-blot hybridization併用法で変異を測定した。その変異頻度とプロテアーゼの活性レベルが逆相関する派生株からRNAを採取し、RT-PCR mRNA Differential Display法を用い、発現が誘導あるいは抑止される遺伝子を検出した所、変異誘導制御に細胞接着分子であるsyndecan-1の遺伝子の関与が示唆された。
