S期特異的に発現する核蛋白質の新しい遺伝子の単離と放射線に対する応答に関する研究

研究概要	S期特異的に発現する新規核蛋白質NP95のcDNAクローンを分離した。この遺伝子産物は、95kDaの分子量を持ち、放射線発がん高発系B10系マウスでは前がん細胞の発生する時期に高発現することや、リンパ腫では細胞周期によらず異常発現する。また抗Np95(Th-10a mAb)をFITC標識し、抗PCNA(PC10)をTexas Redで標識して、m5s細胞を二重染色して解析したところ、Gl期及びS期において核内でNp95とPCNAの局在が、ドット状に一致して分布していることが示された。またmetaphaseやanaphaseの細胞分裂の各時期においても、染色体のまわりに結合している像が観察された。平成10年度においては、Np95の遺伝子のターゲッテングベクターを作成し、ES細胞に導入し、ノックアウトマウスの作成することを試みた。その結果、ES細胞にNp95の遺伝子のターゲッテングベクターを組み込むことに成功し、さらにネオマイシンの濃度を上げることによって、もう一方の対立遺伝子のNp95遺伝子も欠損している細胞株ES.Np95-I-を作成することが出来た。現在、ES.Np95+/-細胞を、マウスのBlastocystに入れて、ノックアウトマウスを作成中である。またNp95遺伝子欠損ES.Np95-/-細胞について、放射線に対する反応性を調べたところ、放射線感受性であることが、明らかになった。これらの結果から、Np95は、DNA修復、複製、細胞周期に関与する調節蛋白の可能性が示唆される。今後は、UVやアルキル化剤に対する反応やアポトーシスとの関係を解析し、Np95の機能解析をさらに進める。
