DNA結合性タンパク質HMGIおよびHMG2の機能とその作用機序の解明

研究概要

[1] HMGタンパク質のDNA結合機構とそれに伴う構造変動の解析
HMG1の2つのHMGボックスは単独でもDNA結合能を有すが、隣接塩基性領域の存在と2つのボックスの繋がりが結合安定性を増すこと、強いDNA結合は主にボックスB内の102Fのインターカレーションによることを解明した。また、2つのボックスがhead-to-headに繋がっており、DNAの17から18bpに結合、95から137度のDNA湾曲を与えるものと推定した。以上をもとに、HMG1とDNAと結合機構モデルを提唱した。HMG1/2は、負の超らせん型DNAに対する優先的認識、DNA巻き戻し活性、ヌクレアーゼS1消化からのDNA保護活性などの性質をもつが、その機能発現にはいずれもbox Bが必須であるが、十分な活性発現には結合親和性を高める周辺領域を必要とすることを解明した。
[2] HMG1、HMG2の機能、性質を解析し、以下の新知見を得た。
(a) HMG1よる細胞内での転写促進がクロマチンの構造弛緩を伴うこと。
(b) HMG1,2によるDNA bendingの機能領域はボックスBと隣接配列であること。
(c) DNA依存性プロテインキナーゼの調節成分としてHMG1,2が機能すること。
(d) DNAリガーゼIVの反応をHMG1,2が促進、制御因子と働く可能性があること。
(e) HMG1,2が外来遺伝子の宿主ゲノムDNAへの組込みを促進する活性をもつこと。
(f) HMG2が細胞増殖の制御に関わる因子であること。
(g) HMG2の核移行シグナルが新規のシグナルであること
(h) 難病患者血清中に見いだしたHMGに対する自己抗体のエピトープを解明したこと。
以上、HMG1、HMG2は転写、複製、組換え、修復、DNAインテグレーションなどに広く関与する多機能性の核タンパク質であることが、明らかにできた。
