C.elegansゲノムプロジェクトが産出する全ガレクチン遺伝子の機能的解析

研究概要	本研究では、既知ガレクチン(32kDa、16kDa)と類似した11種のガレクチン候補遺伝子をゲノムライブラリーから抽出し、系統的な機能解析を通しガレクチン家系の分子進化やガラクトース認識が線虫で果たす役割について考察した。手順として、1)RT-PCRによる発現量の見積もりとESTデータベースの解析、2)cDNAクローニングによるエキソン・イントロン境界領域、および読み枠アミノ酸配列の決定、3)大腸菌発現系の構築と組み換えタンパク質LEC-1-10の調製、4)LEC-1-11の生化学的諸性質の検証、5)糖結合活性の解析、6)内在生レセプターの検索を行った。その結果、先に単離した32kDaガレクチン(LEC-1)と同様、直列反復型の分子構築様式をしたガレクチンが5つ(LEC-1-5)、プロト型で先の16kDaガレクチン(LEC-6)とは若干様相の異なるガレクチンが5つ(LEC-7-11)存在することが判明した。ただし、lec-7に関しては目下のところ成熟タンパク質の発現は確認できていない(第1イントロンのスプライシングの際、終止コドンが出現)。これら2つのグループは相互の類似性が高い(>40%)ことから近縁のサブファミリーを形成する。中でもLEC-1-3間のアミノ酸配列の一致は70%にも及ぶ。しかし、LEC-3はEST解析によると胚のみで発現しており、そのためmixed stageの虫から調製したRNAを用いたRT-PCRでは発現量がきわめて低い。さらに、LEC-3は糖結合に関わるとされる2本のβストランド(S4/S5)間に12アミノ酸からなる余剰ループの挿入があるなど、他のガレクチンにはない特徴をもつ。LEC-4,5は類似性は低い(約40%)が、ともに第1ドメインS2/F1ストランド間に接着モチーフとして知られる「Arg-Gly-Asp」配列をもつ。LEC-8,10,11はさらにユニークで、C-末端に40-80アミノ酸からなる余分なテールをもち、しかも金属キレートカラムに強く結合する。このように線虫ガレクチン家系が単に増幅の一途をたどるのではなく、様々な性質をもったタンパク質に機能分化している様子が観察された。
