移植片拒絶反応のメカニズム:自己-非自己認識機構の分子生物学的解析

研究概要	種の保存、外部からの侵入微生物の傷害排除、個体内部に発生した異常細胞や癌細胞の検出排除や自己免疫疾患など、自己-非自己認識機構の解明は、生物個体の恒常性維持に関する最も重要な研究課題であるが、そのメカニズムは、意外と明らかではない。我々は、移植非自己がん細胞拒絶時に大量に浸潤しているマクロファージ(Mφ)に注目し、このAllogrart-induced Mφ(AIM)が、移植非自己がん細胞を直接傷害する主たるエフェクター細胞であること、AIMが形態、表面抗原、および機能から、まったく新しいタイプの活性化Mφであることなどを明らかにしてきた。平成9年度は、AIMによる移植片傷害機構を、ノックアウトマウス等を用い分子生物学的に解析した。 C57BL/6マウスの腹腔内にBALB/cマウスで継代した線維肉腫細胞(MethA)を移植すると、移植局所に2種類のcytotoxicな細胞が浸潤した。移植したMethA細胞を傷害する主たる細胞は、Allograft-induced Mφ(AIM)であり、移植後4,5日で移植局所に出現した。本細胞の浸潤から遅れること1〜2日後には、移植したMethA細胞には無効だがdonor-typeリンパ芽球にcytotoxicなT細胞(CTL)が浸潤した。CTLの細胞傷害機構は、パ-フォリンによるが、AIMは、Ca^<2->依存的、パ-フォリン-、Fas-非依存的に移植MethA細胞をアポプト-ティックに傷害することが判明した。これらの結果は、他の研究者による皮膚や臓器(腎、心など)移植での傷害機構に関する実験結果(パ-フォリン-、Fas-非依存的)と非常に良く合う。
