新たに見つかった核酸の2重鎖-4重鎖スイッチングの分子的基盤の解明とその利用

研究概要

d(GGAGGA)がカリウムイオン存在下で形成する新規4重鎖構造及び同DNAがカリウムイオン非存在下で形成する新規2重鎖構造をNMRを用いて原子レベルの分解能で決定した。特に後者においては非標準的塩基対であるG:G及びA:A塩基対のみからなるユニークな構造が明らかになった。
抗ガン剤UCH9とDNAの複合体の構造をNMRにより決定し、UCH9の結合によって引き起されるB型→A型の構造変化及びその生物学的意義を明らかにした。
RNA結合蛋白質hnRNP DOとMusashi1の立体構造をNMRにより決定し、さらにこれらによって引き起される核酸の4重鎖-1重鎖スイッチングをはじめて同定した。この活性とテロメアDNA維持機構との関連についても知見を得た。
