核蛋白質輸送における核膜孔ターゲティング複合体の役割

研究概要	本研究者らは、核蛋白質を細胞質で認識して核膜孔ヘターゲットする58kDa(FTAC58/imponin α)と97kDa(PTAC97/importinβ)の2つの因子を同定し、その機能解析を進めてきた。importin αは、核蛋白質の核局在化シグナル(nuclear localizain signal:NLS)に結合するNLS受容体として機能し、分子ファミリーを形成することがわかった。各importinαファミリー分子は、NLS認識に多様性を示し、NLS受容体として異なる機能を有することが示唆された。一方、importinβはimportinαファミリーを介して多様な核蛋白質を核内に運搬する輸送担体として機能する。importinβはimportinαが認識した核蛋白質を核内に運ぶために3つの重要な相互作用をもつ。867個のアミノ酸からなるこの分子は450番目のアミノ酸からC末にかけての領域でimportinαと結合し、N末から449番目のアミノ酸の領域でGTP型のRanと核膜孔複合体(nuclear pore complex:NPC)構成因子の両方に結合する。Ranとの結合にはN末半分の領域がとる特定の立体構造が必要であり、150450番目のアミノ酸の領域でNPC構成因子と相互作用する。importinβは、そのNPC構成因子と相互作用する領域に依存して核-細胞質問をシャトルする。importinβは、Ranなとの他の細胞質性因子を必要とせずに、自身のNPC構成因子と相互作用する領域にのみ依存して核膜孔を通過することが判明した。importinβの挙動は、RanのGTPase活性が分子の核膜孔通過に必要であるという従来の概念に大きな疑問を投げかけた。importinβのように、それ自身で核膜孔を通過する能力をもつことが、輸送担体の重要な性質として新たに定義できる。Wnt/Winglessシグナル伝達の重要な構成因子であるβ-cateninも、importinβと同様に、Ranを必要とせずに、それ自身で核膜孔通過能をもつ分子であることが判明し、細胞質性因子を利用しない核蛋白質輸送経路が細胞内に存在することを示した。当初の研究目的は十分に達成できたと思われる。
