研究概要	低分子量GTP結合蛋白質Rhoが、アクチン細胞骨格の再編成を介して細胞の接着や運動を始め種々の細胞機能を制御していることが明らかにされている。特に、低分子量GTP結合蛋白質がカスケード的に働いてこれらの細胞機能を制御している可能性が示唆されている。本研究では、出芽酵母の低分子量GTP結合蛋白質カスケードの分子機構を明らかにするために、低分子量GTP結合蛋白質カスケードの最下流で働くRHO1の活性制御機構と作用機構について解析し、以下の成果を得た。 (1)RHO1の活性制御機構:RHO1のGDP/GTP交換反応促進蛋白質であるROM1/ROM2に結合する蛋白質をTwo-hybrid法を用いて検索し、数種類の新しい遺伝子を得ることに成功した。 (2)RHO1の作用機構:RHO1の標的蛋白質であるBNI1に結合する蛋白質を検索してSPA2とElongation Factor 1α(EF1α)を得た。細胞生物学的、生化学的な解析により、SPA2はBNI1を細胞膜につなぎ止めることを、EF1αはBNI1によって制御されてアクチン線維の再編成を制御することを明らかにした。 (3)BNR1の作用機構の解析:BNI1に類似するBNR1(BNI1-Related)を高発現すると細胞質分裂異常を引き起こすことを明らかにした。さらに、BNR1に結合する新しい蛋白質としてHOF1を同定した。HOF1はSH3デメインを持ち、BNR1と同様に細胞質分裂部位に局在し、その変異株は細胞質分裂異常を示した。このように、私は、本年度の研究予定をほぼ達成することができた。
