肥満細胞における脂質性メデイエーター代謝酵素群の機能的連関

研究概要	本研究では、マウス骨髄由来培養肥満細胞(BMMC)、ラット肥満細胞株RBL-1及びラット腹腔肥満細胞(CTMC)を用い、アラキドン酸代謝系及び血小板活性化因子(PAF)代謝系について、各代謝酵素の機能連関を調べた。 1. アラキドン酸代謝系 (1) BMMCをIgE/抗原で刺激した際に惹起される二相性のPGD_2産生について、即時相では常在型酵素であるcPLA_2とCOX-1が、遅発相では誘導型のsPLA_2-IIAとCOX-2が機能連関することを見出した。 (2) ラットCTMCのNGF依存的遅発型PGD2産生におけるCOX-2の重要性を確立し、sPLA_2-IIAがCOX-2の発現誘導そのものを増強することを明らかにした。 (3) sPLA_2-IIA欠損マウスのBMMCを用いた解析により、肥満細胞が新規sPLA_2分子種であるsPLA_2-IICとsPLA_2-Vを発現していることを見出し、このうち後者がsPLA_2-IIAの機能を代替する可能性を提唱した。 (4) BMMCのアポトーシスに伴うアラキドン酸遊離におけるsPLA_2-IIAの役割を明らかにした。 (5) RBL-1細胞を用い、LTC_4合成酵素の誘導機構について検討した。 (6) 繊維芽細胞との共培養系を用いてBMMCをCTMC様の成熟肥満細胞に分化誘導する系を確立し、この細胞を用いてG蛋白依存的刺激に伴うPGD_2産生系の分子機構を解明した。 2. PAF代謝系 (7) 種々の肥満細胞が脱顆粒反応に伴ってPAFアセチルハイドロラーゼを細胞外に放出することを見出した。この酵素の分泌に件って、肥満細胞自身が産生したPAFは速やかに分解・不活化された。また、本酵素の発現がサイトカイン刺激によって誘導されることを初めて見出した。
