卵成熟における蛋白質翻訳開始の分子機構

研究概要

動物卵成熟誘起において重要な意味を持つサイクリンBmRNA翻訳開始の分子機構を明らかにすることを目的に本研究は遂行され、本年度は特に、Pumilio蛋白質に関する研究で成果を得た。Pumilioはショウジョウバエの形態形成に関わる重要な蛋白質の一つで、mRNAに結合し、その翻訳を抑制する機能を持つと推測されている。キンギョやアフリカツメガエルサイクリンB1mRNAの3'非翻訳領域(UTR)にはPumilioが結合する配列に似た配列が存在することから、サイクリンBmRNA翻訳調節にこの蛋白質が関与する可能性がある。本研究ではキンギョとアフリカツメガエルPumilioホモログcDNAをクローニングし、モノクローナル抗体を作製した。まず、得られた抗体によるウエスタンブロッティングにより、キンギョ・ツメガエル未成熟卵及び成熟卵、両方にこの蛋白質が存在することを確認した。続いて、poly(U)Sepharoseとoligo(dT)celluloseにより精製したmRNA結合蛋白質中に、Pumilio蛋白質が存在することを確認した。さらに、RNAアフィニティークロマトグラフィーとノースウエスタンブロッティングにより、Pumilio蛋白質がサイクリンBmRNA、特にその認識配列に類似の配列が存在する3'UTR部に、特異的に結合することを確認した。以上の結果はPumilio蛋白質がサイクリンBmRNA結合蛋白質であることを証明し、サイクリンBmRNA翻訳調節機能を果たしている可能性を強く示唆する。今後、PumilioによるサイクリンBmRNA翻訳抑制とホルモン刺激によるその解除の分子実体に切り込む予定である。
