パーキンソン病ドーパ療法におけるセロトニンニューロンの役割

研究概要	モミアミン酸化酵素(MAO)にはA型とB型のアイソザイムがある。パーキンソン病の有効な治療法としてドーパ療法がある。投与されたドーパは線条体においてドーパミンに変換され、欠乏しているドーパミンを補うと考えられている。さらにドーパとともにB型MAOの阻害剤であるデプレニールを合わせて投与すると、ドーパの治療効果を増大させる。ドーパ療法におけるデプレニールの作用機序を理解するためには、脳におけるB型MAOの分布を明らかにする必要がある。本研究では、酵素組織化学を用いて、ラット脳のセロトニンニューロンとノルアドレナリンニューロンのMAOの性質を調べた。A型MAOとB型MAOの共通基質であるチラミンを用いると、セロトニンニューロンとノルアドレナリンニューロンにMAO活性が認められた。デプレニールはセロトニンニューロンのMAO活性を阻害したが、ノルアドレナリンニューロンのMAO活性は阻害しなかった。反対に、A型MAOの阻害剤であるクロルジリンは、ノルアドレナリンニューロンの活性を阻害したが、セロトニンニューロンのMAO活性は阻害しなかった。さらに、A型MAOの特異的基質であるセロトニンを用いると、MAO活性はノルアドレナリンニューロンに認められたが、セロトニンニューロンには認められなかった。また、B型MAOの特異的基質であるフェニルエチラミンを用いると、MAO活性はセロトニンニューロンに認められたが、ノルアドレナリンニューロンには認められなかった。以上の結果から、ラット脳のセロトニンニューロンはB型MAOを持ち、ノルアドレナリンニューロンはA型MAOを持つことがわかった。今後はセロトニンニューロンのB型MAOについて、その生理的機能とパーキンソン病ドーパ療法における作用機序を検討していく。
