孤束核神経回路におけるATP受容体を介した神経間情報伝達の機能的役割

研究概要	孤束核においてATP受容体を介した神経間情報伝達が機能しているか否かをスライス・パッチクランプ法を用いて検討した。平成9〜10年度の研究成果から以下の結論を得た。(1)孤束から孤束核2次ニューロンへの神経伝達において、pre側からpost側への伝達物質としてATPおよびATP受容体が用いられている可能性は低い。孤束刺激誘発の後シナプス電流中、CNQX,bicuculline,AP5およびstrychnine抵抗性の成分は検出できなかった。また、従来の急性解離ニューロンを用いた報告とは異なり、ATPによる内向き電流を示す孤束核ニューロンは見い出されなかった。 (2)しかし、孤束刺激誘発の後シナプス電流がa,β-methyleneATPで増強される孤束核2次ニューロンが認められた。さらにATPは同電流の遅延性抑制を起こした。この事実は、従来の形態学的方法でその存在が孤束核内に報告されていたP2X受容体がpost側ではなく孤束1次求心性線維終末に局在し、やはり終末のアデノシン受容体とともにグルタミン酸放出を修飾している新たな可能性を示す。(3)赤外線ビデスマイクロスコピー下に同定された迷走神経背側運動核の大細胞径ニューロンの約70%がATP投与でTTX括抗性の内向き電流を示した。これらのニューロンのすべてはbicucullineによって大部分、CNQXによって一部消失する自発性後シナプス電流を示したが、その頻度および振幅は、ATP投与によって甚だしく増加した。この増加はTTXにより消失した。この事実は、従来、急性解離ニューロンを用いて報告されている副交感神経節前線維細胞のみならす、その活動を抑制性あるいは興奮性に修飾している孤束複合体内神経細胞群にP2X受容体を発現するニューロンが数多く存在し、シナプス連絡を介して諸種内臓機能の制御に関与している可能性を示す。
