前シナプス蛋白質NACPの生理機能:細胞内情報伝達機構への関与

研究概要	我々は、アルツハイマー病(AD)脳アミロイドから未知ペプチドNAC(non-Aβcomponent of AD amylold)を見出し、前駆体蛋白質(NACP)のcDNAをクローニングした。重視すべきは、NACPが多量に発現している部位とAD脳神経病理の分布部位とが酷似することである。ADの老人斑の数と痴呆の程度とは必ずしも一致せず、強い正相関はシナプス数の減少と痴呆の程度とに見られる。NACPの局在が前シナプスという事実から、NACPの変異、機能異常、又は代謝異常がシナプスの変性を来し、そのことが痴呆という症状に繋がる可能性がある。実際、AD脳において、シナプス部位と変性神経突起にNACPが異常に蓄積していることを我々は示した。一方、最近NACP遺伝子の2つのミスセンス変異が、家族性パーキンソン病(PD)の独立した5家系で、疾患原因遺伝子として相次いで同定された。我々はPDの特徴的神経病理であるLewy小体の異常繊維構造にNACP全分子が存在することを免疫電顕で示した。他方、精製した単量体NACPはin vitroで繊維構造を形成する事が示され、変異体は繊維形成が加速した。従って、NACPの異常繊維構造物が細胞内、軸索内、及び前シナプス部に蓄積し、そのことがニューロンの機能異常、つまりは神経伝達異常をもたらし、症状を発現せしめ、神経細胞死にも繋がる可能性が高い。事実、我々は、Lewy小体型痴呆症(DLB)海馬のperforantpathwayの変性終末軸索にNACPが異常構造物として蓄積していることを見出し、他方、軸索の始原ニューロンの胞体には蓄積があまり見られないことから、終末部からの逆行性神経変性を示唆出来た。又、グリア内にもNACPの異常構造体を確認したのみならず、多系統萎縮症の細胞内異常繊維もNACPを構成成分とすることを明らかにし、複数の神経変性疾患の病理形成にNACPが主要な役割を果たしていることが判明した。
