細菌多糖リアーゼの多面的機能・構造相関解析と医薬・食品領域への応用

研究概要	食品多糖(アルギン酸、ゲラン、キサンタン)に作用する多糖リアーゼについて以下の知見を得た。【アルギン酸リアーゼ】緑膿菌は菌体外に多糖アルギン酸(バイオフィルム)を分泌し、重篤な呼吸器疾患を引き起こす。アルギン酸はマンヌロン酸とグルロン酸からなるへテロ多糖である。緑膿菌産生アルギン酸を分解するSphingomonas sp.A1のアルギン酸リアーゼIII(ALYIII)について、その触媒機構を検討した。ALYIIIはアルギン酸をエンド的に分解し、アルギン酸の4糖を認識して結合し、最終的に2糖と3糖にまで分解することが明かとなった。ALYIIIはアルギン酸のマンヌロン酸領域のみならず、マンヌロン酸・グルロン酸からなるへテロ領域にも作用した。【ゲランリアーゼ】Bacillus sp.GL1(GL1)は食品中のゲル化・増粘剤として用いられているゲラン[(G1c-G1cA-G1c-Rha)n]を分解し、菌体外に分解酵素(ゲランリアーゼ)を分泌する。GL1のゲランリアーゼ遺伝子のクローニング及び1次構造の解明を行った。その結果、ゲランリアーゼは、260kDaの巨大な前駆体から翻訳後修飾により140kDaの成熟体になることが判明した。ゲランリアーゼは他のタンパク質と相同性を全く示さず、新規な糖質分解酵素であることか明かとなった。ゲランリアーゼによるゲラン分解物(ゲラン4糖)の生理活性を検討した。ゲラン4糖はヒトの表皮細胞の増殖を促進する機能を有していることが判明し、ヒトの表皮細胞の傷害における治療薬への可能性が示唆された。【キサンタンリアーゼ】GL1は、食品多糖として汎用されているキサンタン(セルロース骨格に側鎖3糖が付加)をも分解し、菌体外に分解酵素(キサンタンリアーゼ)を分泌することが判った。キサンタンリアーゼを精製した結果、分子量75kDaの単純タンパク質であり、キサンタンに特異的に作用した。キサンタンリアーゼは側鎖の末端糖に作用するユニークな酵素であることも明らかとなった。
