細菌多糖リアーゼの多面的機能・構造相関解析と医薬・食品領域への応用

研究概要	多糖(アルギン酸、ゲラン、キサンタン)に作用する多糖リアーゼについて以下の知見を得た。【アルギン酸リアーゼ】緑膿菌は菌体外に多糖アルギン酸を分泌し、重篤な呼吸器疾患を引き起こす。緑膿菌産生アルギン酸を分解するSphingomonas sp.A1のアルギン酸リアーゼIII(A1-III)について、そのX線結晶構造解析と医薬への応用を検討した。A1-IIIの結晶構造を分解能1.8Åで決定した結果、本酵素は大小12個のα-へリックス構造からのみ形成されていた。また、高分子基質(アルギン酸)が貫通しながら酵素の触媒部位と相互作用するクレフトの存在が認められた。医薬への応用にあたり、本酵素の抗原エピトープを一次構造からN末端側Ser27-Cys44に決定した。その領域は立体構造的にも表面に露出していたため、その領域を除去した改良酵素を大腸菌で大量発現させた。現在、改良酵素の触媒能及び抗原性について解析している。【ゲランリアーゼ】Bacillus sp. GLl(GLl)は、食品中のゲル化・増粘剤として用いられているゲラン[(Glc-GlcA-Glc-Rha)n]を菌体外のゲランリアーゼで4糖にまで低分子化した後、3種の菌体内エキソグリコシダーゼで単糖にまで分解した。ゲラン分解に関わる全酵素の諸性質を明らかにし、またそれらの遺伝子のクローニングを行った。分解酵素の一つである不飽和グルクロニダーゼは、不飽和糖質にのみ作用する新規な酵素であった。現在、各分解酵素によるゲランオリゴ糖についてその生理機能を解析している。【キサンタンリアーゼ】GLlは、菌体外のキサンタンリアーゼとグルカナーゼにより、食品多糖キサンタンをピルビルマンノースと4糖に低分子化した後、3種の菌体内エキソグリコシダーゼで単糖にまで分解した。キサンタンリアーゼは、95kDaの前駆体から75kDaの成熟体にプロセシングされることが判明した。キサンタンをリアーゼで処理した修飾キサンタンは、他の多糖(グアーガム)との相乗性が認められ、未修飾体と異なる物性を示した。
