線虫C.elegansのCu^<2+>ポンプATPaseの構造と機能に関する研究

研究概要

(1)C.elegans Cu^<2+>ポンプATPaseの生化学的な機能解析のため、酵母を用いて本cDNAの発現系を確立した。PCRの手法を用いて本酵素にアミノ酸置換を施し酵母で発現させ、機能に必須であるアミノ酸配列(Cys-Pro-Cys、Asp-Lys-Thr-Gly)を同定した。さらに、ヒトのCu^<2+>ポンプATPase(Wilson病原因遺伝子)についても同様の系で機能を解析した。
(2)Cu^<2+>ポンプATPaseのC.elegansにおける役割を調べるため、Cu^<2+>ポンプATPase遺伝子にトランスポゾンが挿入された株をスクリーニングし、Cu^<2+>ポンプATPase欠損変異株を作製した。現在、得られた欠損変異体に、発生の異常、あるいは重金属イオンの代謝異常などが起こらないかどうかを明らかにしている。また同時に、野生体の生育が重金属イオンの存在で影響されるかどうか調べたところ、20μMの銅、カドミウムイオンで顕著な生育阻害があることを見いだした。
(3)C.elegans Cu^<2+>ポンプATPaseの発現様式を調べるため、先ずそのゲノムDNAをクローン化した。本遺伝子は第3番染色体上にあり、その5′上流には重金属応答性のプロモーターが存在していた。次に、その領域をβ-galactosidaseおよびgreen fluorescent proteinをコードする遺伝子と融合し、C.elegansに導入し、レポーターとして用いて本酵素を特異的に発現している細胞を検索した。本酵素は主に虫体先端部の咽頭筋に発現していることを明らかにした。咽頭は直接外部と接触していることから、本酵素が外部からの重金属イオンの取り込みあるいは吐き出しに関わっていると考えている。
