研究概要	昨年度、ヒトを含めた霊長類の精子形成が他の動物と異なり高度に制御されたものであることを神経型ユビキチンC末端水解酵素(neuronalUCH=PGP9.5)を中心に説明してきた。精子形成の始まりでその律速段階である精祖細胞の増殖調節機構にはneuronalUCHが深く関与している。つまり精祖細胞細胞質においてサイクリンやサイクリン依存性リン酸化酵素の分解はユビキチンを介するextra-lysozomal protein degradationによって行われ細胞周期が回転するが、静止状態であるG0期の細胞質にはneuronalUCHが多量に存在し上記細胞周期関連蛋白質の分解を可逆的に抑制している。このneuronalUCHの精祖細胞での発現と細胞質への分布が霊長類精子生産量を決定している。本年度はニホンザル精祖細胞内neuronalUCHの発現と細胞質への分布を調節する因子を検討してきた。まず精祖細胞増殖能および精子生産量に明らかに差のある繁殖期と非繁殖期の精巣の超微形態の観察から精祖細胞が直接接する基底膜に大きな差を見出した。精祖細胞の細胞増殖が強く抑制された非繁殖期には基底膜が把厚し精細管内腔側に大きく貫入していた。これは精祖細胞と基底膜の接触面積が増大したことを示している。そこで基底膜を構成する接着分子のひとつラミニンに注目し免疫電顕を行った。その結果非繁殖期基底膜の内腔貫入部にラミニンが集積したいた。上記結果は第38回日本組織細胞化学会総会で発表し、Acta Histochem Cytochemに推薦された。霊長類精子形成には視床下部・下垂体・性腺系のホルモンを介した調節以外に局所での調節系が存在することを強く示唆できた。
