内皮由来過分極因子の作用メカニズムの解明

研究概要

モルモット冠動脈の摘出標本を用いて、アセチルコリン(ACh)により遊離される内皮由来過分極因子(endothclium-derived hypcrpolarizing factor:EDHF)による血管弛緩反応についての解析を行った。NO合成酵素阻害剤であるN_ω-nitro-L-arginineおよびシクロオキシゲナーゼ阻害剤であるindomethacin存在下で、AChは内皮依存性の弛緩反応を惹起した。この弛緩反応は、非選択的K^+channel阻害薬である4-aminopyridineによって完全に抑制された。一方、large-conductance Ca^<2+>-activated K^+channel(BK channel)の阻害薬であるiberiotoxin、small-conductance Ca^<2+>-activated K^+channel(SK channel)の阻害薬であるapaminおよびATP-sensitive K^+channel(K_<ATP> channel)の阻害薬であるglybenclamideによっては抑制されなかったことから、BK channel、SK channel、K_<ATP> channel以外のK^+channelの活性化による膜の過分極がEDHFによる弛緩反応に関係していると推察された。興味深いことに、BK channelの阻害薬として知られているcharybdotoxinは、EDHFによる弛緩反応を部分的に抑制した。さらに、charybdotoxinとapaminの両方が存在すると、EDHFによる弛緩反応は完全に消失した。以上の結果から、EDHFはcharybdotoxinとapaminの共存により抑制されるK^+channelを活性化することにより、血管弛緩反応を惹起することが明らかとなった。
