変異型Ret蛋白による細胞癌化に特異的なシグナル伝達の解析

研究概要	Retはプロトオンコジーンで、受容体型チロシンキナーゼをコードしており、遺伝性の造腫瘍性疾患である多発性内分泌腫瘍症(MEN)2A型、2B型、家族性甲状腺髄様癌(FMTC)の原因遺伝子である。これらの疾患では、Retの点突然変異によりアミノ酸が置換されてRetの活性化が起きる。また、RetにはGDNF、Neurturinなどの複数のリガンドの存在が知られている。今回、Retの活性化により癌が発生するメカニズムを解明するため、Retの活性化による細胞内シグナル伝達の解析を行った。リガンドにより活性化され自己リン酸化したRetとSH2ドメインを有するアダプター蛋白との結合をスクリーニングしたところ、in vitroでGrb7、Shc、PI3-kinase subunit、Crkとの結合があり、in vivoではShc、Crkとの結合があった。Retの活性化により生じるCrkとRetの複合体には、チロシンリン酸化されたp130^<Cas>、Paxillinが結合していた。またFAKのリン酸化も検出された。これらの蛋白は接着斑関連の分子として知られる。これらの分子のリン酸化とPI3-kinase、Rhoとの関連を調べた結果、いずれもPI3-kinaseの下流であるが、更にFAKのリン酸化はRho依存性で、p130^<Cas>、Paxillinのリン酸化はRho非依存性であることが分かった。癌に見られる浸潤は、細胞の運動性の亢進として理解されるが、Retの活性化による発癌には、上記のメカニズムによる細胞接着の変化が関与している可能性が考えられた。
