膿胸リンパ腫発生におけるEBV関連免疫応答不全の解析

研究概要

1. リンパ腫組織においてEpstein-Barr virus(EBV)の潜伏感染を証明できた16例の膿胸リンパ腫(PAL)症例についてPCR-SSPによるHLA-A alleleのgenotypingを行ったところ、その各alleleの頻度は日本人正常populationとの間に差を認めなかった(manuscript submitted)。
2. 我々の樹立したPAL細胞株におけるEBNA蛋白の発現をWestern blot法で確認し、RT-PCR法にてEBNA4mRNAの発現を認めた。 新鮮凍結組織が保存されていたPAL症例6例についてEBNA4mRNAの発現を検討したところ、4例でその発現を確認した。 さらにリンパ腫組織中のEBV genomeのEBNA4抗原決定基領域をsequencingしたところ、typeAgenomeを有する4例中3例にCTL誘導を減弱させることが知られている変異を認めた。 またtype B genomeを有する3例においてはprototypetype Bと同一の塩基配列であったがprototype typeBそのもののCTL誘導能は弱いことが知られている(manuscript submitted)。 また、患者末梢血中のEBV genomeの同領域の検索では、リンパ腫組織中のEBVのそれとは異なる場合が多く、腫瘍発生過程でEBV genomeの選択が起きていることが示された。
3. EBNA2-6の発現が見られないEBV陽性鼻腔リンパ腫組織中のEBV genomeの同領域を検索したところ、全例typeAであり、10例中1例に変異を認めた。 患者末梢血中のEBV genomeの同領域の検索でも全例typeAであり、ほぼ変異は認められず、末梢血中とリンパ腫組織中で塩基配列に違いを認めない場合がほとんどであった。 PALの場合とは明らかに異なり、PALにおけるEBVの選択は、その抗原性の違いによる可能性が強く示唆された(manuscript submitted)。
