骨組織由来腫瘍細胞誘引因子の単離・同定-ラット骨肉腫細胞株を用いて

研究概要

我々は平成9年度間に、骨形成能を有する細胞に由来する腎細胞癌細胞誘引物質の単離同定を、以下のように行った。
骨形成性ラット骨肉腫培養細胞株YROS-1を無血清培地中で培養し、培養上清を濃縮、Boyden chamber assayにて腎細胞癌細胞株3株の遊走促進活性を検討した。遊走能を促進する活性を有する分画を各種カラムを用いて分離した。得られた分画はCoumassie blue染色にて還元条件下で40kD,非還元状態で35kDの単一バンドを得た。
分子量ならびに骨芽細胞系由来であることを鑑みて、SPARC/Osteonectin/BM-40が活性物質であると仮定した。ウエスタンプロットにて同バンドは、抗SPARC抗体と反応性を示した。抗SPARC抗体を処理することによって遊走促進活性は失われた。
さらに、Boyden chamberのコート基質をtype IV collagenから、fibronectin,laminin,などに変えたところ、遊走促進能は低下を示した。
SPARCは骨のみでなく、血管内皮細胞、血小板に広く存在していることから、骨に限らず、血管壁に対する腫瘍細胞の遊走能を促進させている可能性が示唆された。
さらに、多くの腫瘍細胞株でSPARCの産生が見られており、腫瘍細胞が分泌するSPARCが、type IV collagenと結合することによって自身の浸潤を促進している可能性も示唆された。
今後は、生体内での研究が必要と思われ、現在SPARC欠失マウスを用いての検討を計画中である。
