研究概要	インターロイキン(IL)15は、T細胞の活性化、増殖を促すIL-2様の活性を持つサイトカインとして報告された。特にIL-15はIL-2と異なり多くの組織にmRNAの発現が認められるため、各臓器や局所における免疫応答に深く関与する可能性が考えられる。ところが、IL-15遺伝子の発現とタンパク質としての産生が必ずしも一致しないことが報告された。我々は、マウスIL-15のプロモーター領域を解析したところ、TATAボックスをもたないGCボックス依存的ハウスキーピング遺伝子であることを明らかにした。このことから、IL-15のタンパク質レベルでの産生は、mRNAへの転写後翻訳削飾によるものと考えられた。さらに、活性化マクロファージからIL-15cDNAをクローニングしたところ、数種類の選択スプライシングの結果生じたcDNAが認められた。それらのうち、イントロン部分の配列を含むcDNAを見い出し、新しいエクソンΦの存在を明らかにした。それぞれのスプライシング産物の翻訳効率を調べると、新しいエクソンΦの挿入によって著しい翻訳効率の上昇が認められた。これらのことから、IL-15のタンパク質の産生には新しいエクソンΦのスプライシングによる後転写翻訳調節機構が存在することを明らかにした。
