HDLによる泡沫細胞コレステロールの引き抜き制御機構 -細胞膜アポリタンパク質A-I受容体の同定-

研究概要	【1】アポ蛋白A-I依存性コレステロール引き抜き実験系の確立アセチルLDL(アセチルLDL)を放射標織コレステロールエステル(CE)によってラベルし、これをマクロファージ(Mφ)と保温して、泡沫細胞のCEを標識した。一方、HDLを脱脂後、ゲル濾過によってアポ蛋白A-Iを精製し、上記の放射標識泡沫細胞と反応させ、培地中へ放出されるコレステロールの放射活性を測定した。その結果、アポ蛋白A-Iの濃度依存的、又、反応時間依存的な培地中の放射活性の増加が観察された。本系を【2】のスクリーニングにもちいた。【2】アポ蛋白A-Iによるコレステロール引き抜き作用を阻害するモノクローナル抗体の分離マウス腹腔Mφの細胞膜画分をラットに免疫し、抗体価の上昇を確認した後、脾臓をマウス由来のミエローマ細胞株と融合させた。HAT培地中で融合細胞を選択し、その培養上清を上記のアッセイ系に添加して、効果を検討した。一回目のスクリーニングでは、アポ蛋白A-Iによるコレステロール引き抜き作用を阻害する抗体の取得には至っていない。【3】HepG2由来アポ蛋白A-I含有リポ蛋白の抗動脈硬化作用の検討ヒト肝癌由来株HepG2細胞の培養上清から、幼若HDLのモデルとされるアポ蛋白A-I含有リボ蛋白(HepG2-HDL)を分離し、そのin vitroでの抗動脈硬化作用を検討した。アセチルLDLをHepG2-HDLと無細胞系で保温後、ゲル濾過によって再分離し(再分離アセチルLDL)、そのMφによる分解活性並びに泡沫化活性を検討したところ、HDLと保温したコントロールに比較してそれぞれ20%程度減少していた。幼若HDLの抗動脈硬化作用は、コレステロール引き抜き作用に加え、化学修飾LDLのMφへの取り込み活性の減弱作用によることが考えられた(Biochemistry 36:9816-9825,1997)。
