脳・中枢神経系に発現する甲状腺ホルモン代謝酵素の構造及び発現調節機序の解明

研究概要

甲状腺より主に分泌される主な甲状腺ホルモンであるthyroxine(T4)は、prohormoneであると考えられ、代謝酵素であるiodothyronine deiodinaseにより、3,5,3'-triiodothyronine(T3)に変換されてその作用を発現する。T4よりT3への転換を触媒するdeiodinaseは、臓器分布や生化学特性の違いにより、Type1及びType2の2種類のサブタイプが存在する。我々は、ラット脳cDNA Iibraryより、ラットType2deiodinase cDNA全長約〜5kbを単離し報告した(論文1)。更に、培養ダリア細胞をもちいて、Type2 deioninaseの発現調節の研究を行い、従来報告されてきた、cAMPの刺激のみならず、CGMPの刺激によっても、Type2 deiodinase mRNAレベルが増加する事、CGMPをsecond messengerとするatrial natriuretic peptideや、C-type natriureticpeptideによってもType2 deiodinase活性が増加する事を報告した(論文1)。更に、培養ダリア細胞に、ニコチンを添加する事により、Type2 deiodinase活性が増加する事を見いだし報告した(論文2)。一方、従来より、実験的糖尿病ラットにおいては、血中T3値が低下し、肝臓、腎臓のType1 deiodinase活性が低下する事が報告されている。我々は、糖尿病状態におけるType1 deiodinase活性の低下が、主に翻訳前のレベルで来している事を見いだし報告した(論文3)。更に、肝臓、腎臓におけるType1 deiodinaseの発現が、他の多くの系のマウスの約10%程度しかない遺伝的Type1 deiodinase低発現マウスであるC3IIマウスにおいても、実験的に糖尿病状態とする事により、肝臓、腎臓のType1 deiodinaseの発現が更に低下する事を見いだし報告した(論文4)。更に、ヒト末梢単核球より、Type1 deiodinase mRNAレベルをRT-PCR法により測定する方法を確立し、バセドウ病患者の末梢単核球のType1 deiodinase mRNAレベルは正常健康人のそれと比べて有意に高値である事を報告した(論文5)。
