白血病細胞の新しいアポトーシス誘導因子の精製とその遺伝子クローニング及び臨床応用

研究概要	白血病株細胞の分化誘導時の培養上清中にアポトーシスを促進する活性が存在することを見出し、精製を行った。そのN末端アミノ酸配列決定し、内皮細胞由来インターロイキン8(IL-8)と同定された。事実、組み換え型内皮細胞由来IL-8は各種白血病細胞株(K562,HL-60,Jurkat,THP-1,U937,KG-1など)に対してアポトーシス誘導活性を示した。一方、組み換え型単球由来IL-8にはアポトーシス誘導活は認めなかった。内皮細胞型IL-8の生体内での生物学的意義を追求するため、正常臍帯内皮細胞を用いた研究では、IL-1やTNFによる刺激のほかに、白血病細胞が内皮細胞と相互作用するだけで、内皮細胞は内皮細胞型IL-8を放出することがわかった。そのIL-8は白血病細胞に対してアポトーシス誘導効果を示した。このことから、今まで血管内外の支持細胞としてだけの存在であった内皮細胞が、内皮細胞型IL-8の放出による抗腫瘍効果を有することを新しく意義づけた。さらにBalb/cヌードマウスの腹腔内および皮下の白血病細胞に対する内皮細胞型IL-8のin vivoにおける抗腫瘍効果について検討した。コントロールとして生食投与群、単球型IL-8投与群、TNF投与群を用いた。内皮細胞型IL-8投与群ではTNF投与群とほぼ同程度に腹腔内および皮下の白血病細胞の増殖抑制がみられ、それは腫瘍細胞へのアポトーシス誘導効果によるものであった。一方、生食投与群、単球型IL-8投与群ではアポトーシス誘導効果は認められず、腫瘍の抑制効果が認められなかった。以上より内皮細胞型IL-8は生体内でも抗腫瘍効果をみとめ、その作用機序の解析ならびに新しい白血病治療の開発など、将来臨床応用へと進展させる可能性が示唆された。
