β_3アドレナリン受容体遺伝子多型による成人病発症予知の有用性の検討

研究概要	【目的】β3アドレナリン受容体(AR)は、新生児の褐色細胞や成人の内臓脂肪に多く認められ、エネルギー消費や体脂肪分布に関与するといわれている。1995年にはβ3AR遺伝子異常が、肥満、糖尿病の早期発症などに関与する報告があり。(1)平成9年度は、β3AR agonist(ARa)投与ラットモデルを作成した。今年度は、卵巣摘出ラットにおける肥満に対する摂食制限及びβ3ARa効果、さらにレプチンや骨代謝に関する影響について追加検討する。(2)日本人妊婦(126妊娠)におけるβ3ARのTrp64Arg変異の妊娠分娩におよぼす影響について検討する。【成績】(1)(1)エストロゲン欠乏により、自由摂食下では体重増加、腹腔内脂肪(VF)増加、総コレステロール、中性脂肪(TG)増加をもたらすが、摂食制限により抑制できる。(2)ARaは体重減少をおこさずにVF、TG減少をおこす。(3)血清レプチン値には、エストロゲン量よりもVFが関与していることが判明した。(4)骨代謝への影響については、現在骨薄切標本作成し、骨形態計測中である。(2)日本人におけるβ3ARのTrp64Argヘテロ変異妊婦は、Wildタイプ妊婦に比べ、妊娠時体重増加が大きいのみでなく、新生児体重や胎盤重量がより大きいことが判明した。これらの研究から得られた結果は、平成11年4月の日本産科婦人科学会学術集会、7月の日本骨代謝学会、10月の国際閉経学会などで発表し、論文報告する予定である。今後は、さらに日本人のデータを集積し、β3ARの遺伝子多型が、将来の生活習慣病のスクリーニングとして有用かどうかを検討したい。
