培養シュワン細胞移植による損傷視神経の再生促進手段の開発

研究概要	変性過程に陥った視神経は再生不能であると考えられてきたが、末梢神経由来の培養シュワン細胞に種々の操作を加えて人工移植片を作成・移植し、人為的な環境によって視神経の再生が誘導可能であるかを試みた.新生仔ラット脊髄後根神経節よりシュワン細胞を単離・培養し、細胞数約10^6/mlとなるようにLaminine,Fibronectin,type4 collagen等の細胞外基質を含むMatrigelを80%、さらに神経成長因子であるBDNFを50ng/ml濃度にて調整し、内径1mmのシリコンチューブへの充填によって人工移植片を作成した.成熟ラット(Wistar種、200-300g)の視神経を眼窩内で切断し人工移植片を眼球側視神経に移植し、遠位端は生体接着剤を用いて盲端とした.術後2週および3週において、ロ-ダミンによって順行性に標識された視神経線維が人工移植片内に伸長しているのが確認された.共焦点レーザー顕微鏡を用いた免疫組織化学の観察において、人工移植片内の神経線維にGAP43の局在を認め、L1,NCAMの細胞接着因子を発現していることがneurofilamentとの2重染色によって示された.またミエリン蛋白であるMAGに関しては、術後2週目以降において陽性線維が人工移植片内に確認された.電子顕微鏡では移植片内のほとんどの再生線維がシュワン細胞と接触しており、細胞外基質成分であるMatrigelのなかを伸長する像は認められなかった.移植片内DiI投与による網膜神経節細胞の逆行性標識において約30%の生存・再生率を認めた.これらの結果より、培養シュワン細胞を用いて人為的に作成した人工移植片の環境であっても視神経の再生が誘導可能であること、また末梢神経移植による再生時同様、シュワン細胞を足場として伸長しており、L1,NCAM等の細胞接着因子の発現が見られた.また再生線維においてMAGの発現もあることから、人為的環境内であってもミエリン化が起きる可能性が示唆された.
