コンドロトランスフェリン遺伝子のクローニングと発現制御

研究概要

クローニングに成功したCTF遺伝子の転写開始点を決定するために、BALB/cマウス胎生17日胎児よりmRNAを抽出し、プライマー伸長法を行った。この結果、翻訳開始コドンATGから159bp上流の位置が転写開始点であることが判明した。
さらに、5'上流領域を約2.5キロベースほどシークエンスし、塩基配列を決定した.その結果、CTF遺伝子にはTATAボックスは見出されず、GCボックス様配列、CAATTボックス様配列は存在していた。また、転写因子結合部位データベースでのホモロジー検索を行ったところ、CTF遺伝子の5'上流領域の配列では、AP1やSOX9といった転写因子の結合部位様配列およびCRE、オクタマ-といったコンセンスシークエンス様配列が見出された。これらのいずれかが調節エレメントとして機能し、CTF遺伝子の発現制御が行われている可能性が示唆された。
また、軟骨に誘導可能なマウスATDC5細胞から2,6,10,14,18日にRNAを抽出し、RT-PCR法にてマウスコンドロトランスフェリン(CTF)の遺伝子発現を検討した。CTF遺伝子の発現パターンは同時に行った軟骨細胞の分化マーカーであるII型コラーゲン遺伝子と同様なパターンを示し、軟骨組織で特に大量発現しているCTFは、軟骨細胞の分化における新たなマーカーになるうることが示唆された。
現在、軟骨に誘導可能なマウスATDC5細胞などを用いたルシュフェラーゼアッセイによる、CTF遺伝子のプロモーター活性の検討を試みているところである。今後、CTF遺伝子のさらなる上流解析を通して、軟骨特異的な転写調節を探求してゆく。
