ビタミンによる耳下腺アミラーゼ分泌の脱感作現象とその分子機構

研究概要

近年、中高年者を中心に唾液分泌障害に起因する口腔機能障害を訴える患者が急増しており、この病因の追求と治療法の開発が重要視されている。私はラット耳下腺を用いて、ヒスタミン(H)の短時間の前処理によって誘導されるアミラーゼ(A)分泌の脱感作現象を実験系として、この現象の誘導機構をH_2受容体を介する細胞内情報伝達系を中心に追究し、下記の成果を得た。
(1) H受容体の動態
HはH_2受容体に作用してA分泌を惹起した。[^3H]チオチジンを用いた結合実験により、Hの前処理によるH_2の受容体の減量とH_2作動薬に対する親和性の低下が認められた。また、抗H_2受容抗体を用いたイムノブロット解析でもH_2受容体の減量が認められた。
(2) アデニル酸シクラーゼ(AC)活性の変動
組織内c-AMP量はH前処理によって減量した。従ってH前処理により誘導される脱感作現象にAC活性の低下が示された。
(3) GTP結合(G)蛋白質の動態
H前処理によって促進性G蛋白質(Gs)ではなく、抑制性G蛋白質(G_1)のADP-リボルシル化が促進された。G蛋白質の機能はこの蛋白質のりん酸化により調節され、Giの機能はc-AMP依存性プロティンキナーゼによるりん酸化によって抑制され、プロティンホスファターゼ(PP)2Aによるりん酸化によって増強された。この脱感作現象の誘導にはPP2A活性上昇によるGi2αの脱りん酸化が連関し、オカダ酸によってPP2A活性を阻害すると、この現象は完全に解消された。しかし、免疫抗体を用いた実験において、Gi2αやGsαの量的な変動は認められなかった。
以上の結果からH前処理によるA分泌の脱感作現象の誘導にはH_2受容体の減量及び作動薬に対する親和性の低下と、この受容体に共役しているGi2αの脱りん酸化による機能の亢進が連関していることが明らかにされた。
