赤潮形成種を中心とした渦鞭毛藻の内因性増殖促進物質

研究概要

私は微細藻類の内因性増殖促進因子について研究を進めている。我々は以前、有害プランクトンである渦鞭毛藻Gambierdiscus toxicusが内因性増殖促進因子を生産していること、およびG.toxicusが生産するポリエーテル化合物のガンビエル酸がG.toxicus自身に対して増殖促進作用を有することを報告した。今回、さらに研究を進めポリエーテル化合物を幅広く認識するシガトキシン抗体を使用した実験結果から、G.toxicusにとってポリエーテル系の内因性増殖促進因子は良好な生育のために必須なことが明らかとなった。この知見は8th International Conference on Harmful Algae(Vigo Spain 1997)においても報告しました。またさらにガンビエル酸を生産していないG.toxicusクローンにおいてもポリエーテル物質と考えられる内因性増殖因子の存在を確認し、現在その活性本体についての研究を進めている。
G.toxicusはハワイ、ワイアナエにおいて多種類の海藻が繁茂しているのにも関わらず、緑藻Bryopsis sp.上に集中して見出される。そこでG.toxicusとBryopsis sp.間に何らかの化学物質が介在した関係があることを疑い、Bryopsis sp.抽出物をケイ酸カラムで分画後、各フラクションのG.toxicusに対する作用を調べた。その結果、あるフラクションはG.toxicusを脱皮させ、あるフラクションは底性付着性であるG.toxicusを強力に遊泳させる作用があることを見出した。G.toxicusを脱皮させる成分は、G.toxicus脱皮作用をアッセイとして用い単離することができた。各種分光学的手法による解析から本活性物質は1-palmitoyl-3-O-(6'-sulpho-α-D-quinovopyranosyl)glycerolであることが明らかとなった。(現在投稿準備中)。G.toxicusの遊泳惹起成分については現在研究を進めている。
