トロンボポエチンによる多倍体化を伴う巨核球特異的な分化成熟過程の制御秩序

研究概要	巨核球/血小板の成熟過程には他の血球系の細胞にはみられない巨核球の多倍体化や、巨核球胞体からの突起形成を経た血小板放出などの興味深い現象が存在する。この過程は巨核球/血小板の増殖分化を促進するサイトカインであるトロンボポエチン(Tpo)により担われている。しかし、これらの分子メカニズムに関しては全く解明されていない。まず、巨核球の多倍体化現象を解明するために、マウス骨髄細胞をTpo存在下で長期培養し多倍体化しつつある巨核球を分離し、様々な細胞周期段階における巨核球の中心体や微小管等の分裂装置の実態、核膜の再構成及び細胞周期に関与する蛋白質の発現を蛍光抗体法により検討した。その結果、従来巨核球の多倍体化は単にmitosisをスキップしDNA複製のみが進行するためと考えられていたが、多数の中心体形成、核膜崩壊及び再構成が観察され、少なくともmitosisには正常に入りprophase、metaphase、anaphase Aと進行することが確認された。従ってanaphase B以降において極間の増大等の異常により、複製された染色体が一つの核膜内に包括され多倍体細胞が形成されると結論した。次に、巨核球/血小板産生を制御するトロンボポエチン(Tpo)の発現と血小板産生制御機構の細胞生物学的、分子生物学的解析を行なった。その結果、Tpoは肝、腎で産生されるばかりではなく、筋、脳などすべての組織で恒常的に発現していることが明らかになった。また、急性血小板減少症や増多症では血中Tpo濃度は血中血小板数に反比例して変化するように観察されるが、各組織でのTpoのmRNA量は変化しなかった。抗血小板抗体をマウスに投与して、急性血小板減少症の状態にし、経時的に血中Tpo濃度、血小板数、骨髄と脾臓中の巨核球数を測定した結果、一過性に血中Tpo濃度が増加し、その後巨核球が増加しはじめるとTpo濃度は減少し、巨核球数が減少するとともに血小板数が増加した。したがって血中Tpo濃度は血小板数だけではなく、Tpoが結合可能な骨髄や脾臓中の巨核球と末梢血血小板のTpoレセプターの総数によって規定されるというメカニズムが考えられた。
