ArgosによるRasシグナルを介した細胞分化制御とアポートーシス誘導

研究概要	Argosはショウジョウバエの発生におけるRasシグナル伝達経路を制御する分泌性蛋白質であり、遺伝学的及び生化学的研究によってEGF受容体の活性化を負に制御することが明らかになっている。ArgosによるEGF受容体活性化の阻害機構の解明を目的として、ArgosがEGF受容体へ結合するかどうか、そしてArgosの結合がEGF受容体にどのような影響を及ぼすのかを生化学的に解析した。得られた結果から、Argosという抑制性のリガンドが結合してEGF受容体の二量体形成を阻害するという新しいメカニズムの存在が明らかになった。ArgosによるRaS/MAPKシグナルの抑制および細胞死誘導の分子機構は完全に解明されたわけではなく、未知の分子が関与している可能性がある。これら未知の分子の同定を目的として、複眼特異的にArgosを過剰発現するトランスジェニックフライ(GMR-argos)の表現型を変化させる変異体をスクリーニングした。約140,000の個体をスクリーニングし、有意な効果を示す変異体を複数分離した。これらの変異体を解析したところ、分離した系統の中にはArgosと共通のシグナル経路を介して細胞死あるいは細胞分化の制御に関与する遺伝子の変異体が含まれているものと考えられた。これらの変異をさらにマッピングし、未知のものについては原因遺伝子をクローニングする予定である。これらの遺伝子の機能を解析することによって、Ras/MAPK経路の制御機構および機能について重要な知見が得られるものと期待している。
